Background:
Vitellogenin(VTG)isoneofthecoreendpointsinscreeningandtestingforendocrinedisruptingchemicalsstandardizedintheOECDGuidelinesforthetestingofchemicalsforestrogenicactivity.Originallybelievedtobeproducedonlyintheliver,severalcelltypeshaverecentlybeenshowntoproduceVTGafterestrogenstimulation,includingthoseoftheepidermalmucosaoffish.
GiventhedestructivenatureoftrADItionalsampling,collectingsuccessivesamplesfromfishhasbeenawidespreadproblem,asforinstanceintheanalysisofvitellogenininductionwhilescreeningandtestingchemicalsubstancesforendocrinedisruptionpotential.Bloodisdifficulttocollect,inparticularwhereverysmallfishareconcerned,orinapproacheswheretheanimalsmustsurvivesampling.Thisisparticularlyimportantinfieldmonitoringinordertoavoidimpactonthepopulationunderinvestigation.
EventhoughtheVTGconcentrationintheskinmucusisanorderofmagnitudelowerthaninbloodserumorinbodyhomogenates(containinglivertissue),theskinmucosaisverywellsuitedasamatrixtodetermineexogenousVTGinductioncausedbyenvironmentalchemicalswithaffinitytotheestrogenreceptors.Mucussamplingwiththeaidofswabsconstitutesanalternativetotraditionalprocedures,avoidinganimalinjurybyusingahighlysensitiveELISAincombinationwithahighqualityextractionbufferandswabsampling,thedeterminationofmucosa-derivedVTGhasthefollowingadvantages:
