ThrombinisoneofthemostpotentplateletagoNISTs.It interactswith2protease-activatedreceptors(PARs)on thesurfaceofhumanplatelets–PAR-1andPAR-4.The expressionprofilesofPARsonplateletsdifferbetween humansandnonprimates.PAR-1isbelievedtobethe primaryreceptorforthrombininhumans.Incontrast, mouseplateletsforexamplelackPAR-1andlargely signalthroughPAR-4inresponsetothrombin.

PAR-4testreagent isstableuntiltheexpirydateprinted ontheviallabelwhenstoredat 2-8°C, 14daysafterreconstitutionwhenstoredat 2-8°C,4weekswhen storedat<-20°Cor24hoursafterthawing.If reconstitutedreagentisnottransferredintomicrotest tubes,theoriginalvialshouldbestoredinanupright position.
MeasurementPrinciple:
PAR-1isbelievedtobetheprimaryplateletreceptorforthrombininhumans.Thisreceptorisactivatedby TRAP-6inMultiplate.Incontrasttohumans,severalanimalspecies(e.g.,mice,dogs)lackPAR-1receptorsandlargelysignalthroughPAR-4inresponsetothrombin.Eveninhumans,plateletPAR-4receptorscanbestimulatedtotriggerplateletactivationandaggregation.InordertoreachasimilarextentofaggregationaswiththeTRAPtest,significantlyhigherconcentrationsofPAR-4agonisthavetobeapplied(645µMforPAR-4comparedto32µMforTRAP-6).
