MeasurementPrinciple:
TheCOLtestleadstoplateletactivationviatheplateletcollagenreceptor.Forsuitableplateletactivation,thereleaseofendogenousarachidonicacidfromtheplateletphospholipidsisnecessary.ThearachidonicacidisthentransformedtotheplateletagonistthromboxaneA2(TXA2)bytheenzymecyclooxygenase.CyclooxygenaseisblockedbyAspirin®.Therefore,theCOLtestissensitivetoAspirin®action.ThismechanismalsoexplainswhytheCOLtestislessspecifictowardstheactionofAspirin®ascomparedtotheASPItest.IntheASPItest,adefinedamountofarachidonicacidisusedastheactivator.However,inCOLtest,plateletsareactivatedthroughendogenousTXA2andotherTXA2independentmechanisms.
