TheTECO®UrinaryCollagenIVELISAisdesignedfortheassayofurinarycollagenIV.Itisasolidphaseone-stepsandwichELISA.CollagenIVinthesampleisboundsimultaneouslybyasolidphasemonoclonalantibodyandamonoclonalantibody-enzymeconjugatedirectedatdifferentantigenicsites.ThisresultsinthecollagenIVmoleculebeingsandwichedbetweenthesolidphaseandenzymelabelledantibodies.Afterremovingunboundenzymelabelledantibodyandsample,theplateisthenincubatedwithenzymesubstrate,resultinginthedevelopmentofacolor.ThecolordevelopedisdirectlyproportionaltotheamountofcollagenIVinthesample.

CollagenIVisatypeofnon-helicalformingcollagenfoundprimarilyisthebasallamina(basementmembrane),anditisconsideredtoconstituteitsbasicframework.UrinarycollagenIVlevelsareelevatedinavarietyofrenalpathologies,particularlydiabeticnephropathy.UrinarycollagenIVissignificantlyhigherinpatientswithnon-insulindependentdiabeticmellitus(NIDDM)thaninnormalsubjectsandurinarycollagenIVlevelscorrelatewiththedepositionofcollagenIVinthekidney.Indiabeticsubjects,urinarycollagenIVissignificantlyincreasedinpatientswithmicroalbuminuriaorovertproteinuriaascomparedtothosewithnormoalbuminuria.Moreover,indiabeticswithnormoalbuminuria,thosewithelevatedurinarycollagenIVwereatanincreasedriskforprogressiontomicroalbuminuria.IntensivetherapyofdiabeticnephropathycanslowthetemporalincreaseinurinarycollagenIVindiabeticsindicatingthepotentialofurinarycollagenIVforstudyingtherenaleffectsofnewtherapies.TheseresultssuggestthatthemeasurementofUrinaryCollagenIVmightprovideausefulbioMarkerforstudyingdiabeticnephropathy.
