MeasurementPrinciple:
Technothrombin® TGAisbasedonmonitoringthefluorescencegeneratedbythrombincleavageofafluorogenicsubstrateovertimeuponactivationofthecoagulationcascadebydifferentconcentrationsoftissuefactor(RA,RB,RCLowandRCHigh)andnegativelychargedphospholipidsinwholebloodorplasma.Fromthechangesinfluorescenceovertime,theconcentrationofthrombin(nM)inthesamplecanbecalculatedusingtherespectivethrombincalibrationcurve.Therateofincreaseinthrombinconcentrationovertimethenallowstocalculategenerationofthrombininthesampleperminuteandtoplotthisvalueovertimeforthewholecoagulationprocess.Thisthenresultsinthevisualizationofthedifferentphasesofclotformation.
Background:
TheTechnothrombin® TGAkitisanassaysystemfordeterminationof thrombingenerationovertimeinplateletpoororplateletrichplasma(PPPor PRP)uponactivationoftheclottingcascadebymicellesofnegativelycharged phospholipidscontainingdifferentamountsofhumantissuefactorandCaCl2.The kitcanbeusedtomonitorhemophiliacsduringinhibitorbypassingtherapy,to monitoranticoagulationtherapy,tocalculateINRvaluesforpatientsandto determinestatesofbleedingdisordersorthrombophiliaaswellastheactivityof circulatingmicroparticles.Thisbroadrangeofapplicationsispossiblebyproviding differenttissuefactorconcentrationsandbymonitoringthewholekineticof thrombingenerationduringinitiation,amplificationanddownregulationofthrombin formation.Technothrombin® TGAisthereforeauniversalassaykitfor analyzingandresearching thefunctionofthehemostaticsystem.
