TheTECO®SerumCollagenIVELISAisdesignedfortheassayofserumcollagenIV.Itisasolidphaseone-stepsandwichELISA.CollagenIVinthesampleisboundsimultaneouslybyasolid-phasemonoclonalantibodyandamonoclonalantibody-enzymeconjugate,eachdirectedatdifferentantigenicsites.ThisresultsinthecollagenIVmoleculebeingsandwichedbetweenthesolidphaseandenzyme-labelledantibodies.Afterremovingunboundenzyme-labelledantibodyandsample,theplateisincubatedwithenzymesubstrate.TheresultantcolordevelopmentisdirectlyproportionaltotheamountofcollagenIVinthesample.

Background:
CollagenIVisatypeofnon-helicalformingcollagenfoundprimarilyisthebasallamina(basementmembrane).Chronicliverdiseasecomprisesanumberofprogressivedisorderswhichculminateinlivercirrhosisandwhicharecharacterizedbyexcessivedepositionofcollagen.
Althoughvarioustypesofcollagen(typeI,III,IV,VandVI)increaseintheliverwiththeprogressionoffibrosis,typeIVcollagen,aconstituentofthebasementmembrane,isparticularlynoteworthyforthefollowingreasons:itsserumlevelcorrelateswithhepaticlevelsofcollagenIV,serumlevelsofcollagenIVfallinresponsetoeffectivetherapy,anditistheearliesttypeofcollagentobesynthesizedduringexperimentalliverinjury.SerumcollagenIVlevelsareelevatedinavarietyofliverdiseases,andhavebeenfoundtobepredictiveoftherapyresponseinHepatitisCinfectionandtobesensitiveindicatorsoftherapyresponseinabstainingalcoholics.
